Fermenting SAMPLE BIO BLOG

FERMENTATION AND SOUR PICKLES

Timeframe: 1-4 weeks

Ceramic crock or food-grade plastic bucket
Plate that fits inside crock or bucket
4-liter jug filled with water, or other weight
Cloth cover
Ingredients (for 4 liters):

1.5 to 2 kilograms unwaxed
cucumbers (small to medium size)
90 milliliters sea salt
3 to 4 heads fresh flowering dill, 45 to 60 milliliters of any form of
dill (fresh or dried leaf or seeds)
2 to 3 heads garlic, peeled
1 handful fresh grape, cherry, oak, and/or
horseradish leaves (if available)
1 pinch black peppercorns
Process:

Rinse cucumbers, taking care to not bruise them, and making sure their blossoms are removed. Scrape off any remains at the blossom end. If you’re using cucumbers that aren’t fresh off the vine that day, soak them for a couple of hours in very cold water to freshen them.
Dissolve sea salt in ½gallon (2 liters) of water to create brine solution. Stir until salt is thoroughly dissolved.
3. Clean the crock, then place at the bottom of it dill, garlic, fresh grape leaves, and a pinch of black peppercorns.
Place cucumbers in the crock.
Pour brine over the cucumbers,place the (clean) plate over them, then weigh it down with a jug filled with water or a boiled rock. If the brine doesn’t cover the weighed-down plate, add more brine mixed at the same ratio of just under 1 tablespoon of salt to each cup of water.
Cover the crock with a cloth to keep out dust and flies and store it in a cool place.
Check the crock every day. Skim any mold from the surface, but don’t worry if you can’t get it all. If there’s mold, be sure to rinse the plate and weight. Taste the pickles after a few days.
Enjoy the pickles as they continue to ferment. Continue to check the crock every day.
Eventually, after one to four weeks (depending on the temperature), the pickles will be fully sour. Continue to enjoy them, moving them to the fridge to slow down fermentation.

THE KEYS TO FERMENTATION
The critical ingredients for the fermentation process are:

Salt (sea salt)
Lack of oxygen
Cool temperature

Salting the food preserves the food and protects it from bacteria, so it doesn't spoil before it ferments. Sea salt is most desirable for that purpose, rather than table salt. (Table salt has been bleached and has had other important minerals removed.)

Once the food is salted, it needs to be kept in a cool place with minimal oxygen.

To keep the contents cool, Koreans have traditionally placed their Kimchi pots in the ground, which stays at 55 degrees year-round. Basements and root cellars are also good. For the rest of us, one author recommended using a small refrigerator at the least cool temperature setting.

The final step is keeping the air out, which allows fermentation to occur. It's a process that only takes place in the absence of oxygen. The fermentation does produce a bit of an odor, but you get used to it.
For a cell, fermentation is a way of getting energy without using oxygen. In general, fermentation involves the breaking down of complex organic substances into simpler ones. The microbial or animal cell obtains energy through glycolysis, splitting a sugar molecule and removing electrons from the molecule. The electrons are then passed to an organic molecule such as pyruvic acid. This results in the formation of a waste product that is excreted from the cell. Waste products formed in this way include ethyl alcohol, butyl alcohol, lactic acid, and acetone--the substances vital to our utilization of fermentation.

During lactic acid fermentation, the electrons released during glycolysis are passed to pyruvic acid to form two molecules of lactic acid. Lactic acid fermentation is carried out by many bacteria, most notably by the lactic acid bacteria used in the production of yogurt, cheese, sauerkraut, and pickles. Some animal cells such as muscle cells can also use fermentation for a quick burst of energy.

Other possibilities include the meetings of interest groups like the California Turtle and Tortoise Club, Audobon Society etc....

You must have PROOF of attendance: A picture of you there, a receipt or ticket stub
You should have pictures of what you observed and some reflections on what you learned. Focus on diversity and adaptations, if a species is endangered speculate why. There is a minimum 20 pictures with 3-5 lines about each. More points for more pictures with reflections.

Each activity is worth 200 pts. There is a 1000 pt maximum.

Eaton Canyon Plant Walks: Second Sunday of each month except July and August.

Meet in front of Eaton Canyon Nature Center at 9:00 a.m. (Top of Lake Ave) Then go on a leisurely walk, about 2 hours, through the native plant garden that surrounds the Center and into the nearby wild areas. The walk is different each time — what's leafing out, flowering, in seed, etc., determines what your leader will talk about — and different leaders bring different points of view.

We will finish the lab tomorrow No computer lab :-( Ask me about plugs before you begin again. I believe you are leaving out a step. That means you must do the computer assignment at home. This link will take you to the floating disk lab which you must prelab for next week and a variation of the lab we are doing it might clarify for some of you.

In this computer simulation assignment, you will be manipulating two variables: light intensity and light wavelength. The amount of ATP produced will change depending upon the set parameters.

Go to "Johnson Explorations: Photosynthesis" located at http://www.mhhe.com/biosci/genbio/biolink/j_explorations/ch09expl.htm . Look over the diagram before clicking on "skip intro" so you have a fair idea what is going on.

Procedure: Your task is to use the simulation to determine how wavelength and intensity affect the rate of photosynthesis (and the production of ATP). Keep in mind you are dealing with two variables, so in order to determine absolutely how each factor affects photosynthesis, you must keep one variable constant while manipulating the other variable. This means you will have two parts

Lab Report must contain the following sections.

1. Introduction: Describe briefly the light dependent reactions and propose a hypothesis to answer the experimental question.

2. Data: Include data tables for the simulation. The tables must show clearly trends resulting from changes of intensity and changes of wavelenth. Multiple data tables would probably be best here. In Microsoft Word, use the "table" function to organize your data.

3. Analysis: A graph showing how the percentage of ATP changed (Y axis) as a result of changes in wavelength and intensity (X axis). Two graphs would be best here. You may use Create A Graph to make your graphs, or use a spreadsheet or graphing program of your choice.

3. Conclusion: Use your data to answer the experimental question. Answer clearly how light wavelength affects the reaction, and how light intensity affects the reactions. Offer an explanation of the results, taking into account the principles of photosynthesis and the light reaction.
